# stands for sharp, and it means that you have to play the black key to the right. For example: F# means you play the black key to the right of the F, G# means you play the black key to the the right of G, and so forth. And if for some reason this doesn't reply properly, this was for PokeGuy ^_^

That's a bit misleading. Sharp means a half step to the right. E# is F and B# is C, neither of which are "black" keys.
